“OBSERVING PROTEIN DYNAMICS AND CONFORMATIONAL CHANGES BY ENSEMBLE AND SINGLE-MOLECULE FLUORESCENCE SPECTROSCOPY” Ø ♦ Speaker : 김소연 박사[한국표준연구원] Ø ♦ Place : Physics Seminar Room (Science Bldg, 3-201) Ø ♦ Date & Time : Nov, 11(Wed) 3:00 ~ 4:00 pm Abstract Recent developments in single-molecule spectroscopy and imaging help to illustrate how each molecule works as a component of the network in real time. I will describe two different biological systems using these techniques,: the dynamics of the bacterial actin homologue, MreB protein in living Caulobacter crescentus cell, and the conformational changes of the chaperonin GroEL and its substrate, the von Hippel-Lindau tumor suppressor protein, VHL. Lastly, I will briefly introduce current efforts to observe how mechanical properties, such as stiffness of an extracellular matrix (ECM) and shear flow, affect the dynamics of membrane type 1matrix metalloproteinase, MT1-MMP in the eukaryotic endothelial cell.
